Proprotein convertases (PCs) are Ca2?-dependent serine proteases. Their primary function is to cleave inactive protein or peptide precursors, converting them into active functional molecules. Seven major members of the PCs family have been identified in mammalian cells, including Furin and PC7. These proteases are mainly used in applications related to inhibitor development, such as enzyme kinetics and biochemical assays, as well as the development of antiviral compounds and drugs targeting the novel coronavirus (SARS-CoV-2).
Furin (Furin Protease): Proprotein Convertase Subtilisin/Kexin Type 3 (PCSK3)
In addition to cellular substrates, furin can activate proteins of numerous bacterial and viral pathogens, including the Spike (S) protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Recombinant human furin is expressed in the human HEK293S cell line, which promotes homologous glycosylation. Active furin is selectively purified using inhibitor-based affinity purification. This product is suitable for enzyme assays, activation of furin substrate proteins, and protein crystallography.
