Labeled nucleotides are a class of critical reagents in life science research and serve as effective tools for detecting and labeling specific nucleic acid sequences in molecular biology studies. Typically, labeled nucleotides are inserted into DNA or RNA sequences via enzymatic reactions to enable detection and analysis. Among non-radioactive labeling substances for DNA/cDNA probes, fluorophores and haptens are the most commonly used.
Nick translation is the method of choice for generating labeled DNA probes (100–500 bp) from genomic DNA, plasmids, BAC clones, and other templates. It relies on the coordinated action of DNase I (which introduces random single-strand breaks called "nicks") and DNA polymerase I. At these "nick" sites, DNA polymerase I incorporates labeled dUTP instead of its natural counterpart, dTTP, thereby producing optimal probes for in situ hybridization.
